FOLIKEL ANTRAL OVARIUM PADA PEMBERIAN ANTIBODI MONOKLONA BOVINE ZONA PELUSIDA SEBAGAI KANDIDAT IMUNOKONTRASEPSI
Abstract
Kepadatan penduduk masih merupakan masalah yang penting bagi negara-negara bekembang seperti Indonesia. Di sisi lain penggunaan metode kontrasepsi yang saat ini diandalkan untuk menangani masalah tersebut memiliki bbeberapa kekurangan yang menyababkan upaya menekan jumlah penduduk kurang optimal. Pengembangan metode kontrasepsi dengan target non hormonal seperti penggunan Monoklonal antibodi bovine ZP3 sebagai kandidat bahan imunokontrasepsi perlu dilakukan. Penelitian ini bertujuan mengetahui pengaruh Mab-bZP3 sebagai kandidat bahan immunokontrasepsi terhadap histologi ovarium yaitu jumlah folikel antral yang normal dan atretik. Penelitian True experimental ini dilakukan terhadap 36 hewan coba yang terdiri dari tiga kelompok kontrol dan tiga kelompok perlakuan, masing-masing dianalisa pada hari ke-5, ke-10, dan ke-20 setelah perlakuan. Kelompok kontrol diberikan suntikan Phospat Buffer Saline 50 μl dan kelompok perlakuan diberi suntikan Mab bZP3 sebanyak 50 μl. Hasil penelitian menunjukkan bahwa tidak terdapat perbedaan yang signifikan rata-rata jumlah folikel antral normal (p-value 0,715) maupun folikel antral atretik (p-value 0,604) antara kelompok kontrol dengan perlakuan. Tidak terdapat perbedaan yang signifikan (p-value 0.071) rata-rata jumlah folikel antral atretik antara waktu pemeriksaan pada masing-masing kelompok kontrol dan perlakuan. Namun, terdapat perbedaan yang signifikan (p-value 0.036) rata-rata jumlah folikel antral normal antar waktu pemeriksaan pada masing-masing kelompok kontrol dan perlakuan. Kelompok kontrol yang diperiksa pada hari ke-5, berbeda signifikan hanya dengan kelompok kontrol maupun kelompok perlakuan yang diperiksa pada hari ke-20. Sedangkan rata-rata jumlah folikel antral normal terendah terdapat pada kelompok kontrol yang diperiksa pada hari ke-20, namun tidak berbeda signifikan dengan kelompok perlakuan yang diperiksa pada hari ke-20. Hasil tersebut mengindikasikan bahwa Mab bZP3 sebagai kandidat bahan imunokontrasepsi tidak berpengaruh terhadap perkembangan folikel, dalam hal regulasi apoptosis dan fungsi reseptor FSH pada sel granulosa. Efek kontrseptif bersifat reversible, yang ditunjukkan dengan tidak adanya perbedaan yang signifikan jumlah folikel antral antara kelompok kontrolndan perlakuan pada hari ke-20.
